The MolYsis Complete5 kit was used to deplete human DNA and isolate microbial DNA from peritoneal fluid and blood samples. After which DNA sequencing was performed using MinION (Oxford Nanopore, UK) for 72 hours. After 20 and 30 minutes, the confident detection threshold was met for Mycobacterium tuberculosis and Brucella melitensis respectively. A high confidence level was achieved after six hours for both pathogens.
In conclusion, where traditional culture could not effectively detect the two microbial pathogens. The use of molecular diagnostics gave a rapid result for both bacterial pathogens.
