Agena® MassArray System
The Agena MassARRAY System is an open platform for genetic analysis based on MALDI-TOF mass spectrometry. It multiplexes up to 50 genetic variants in one reaction and delivers results from sample to data within a single day. The platform handles single nucleotide variants, insertions, deletions, translocations, copy number variation and methylation markers, in a genotyping workflow and an EpiTyper methylation workflow.

The Agena MassARRAY System separates and identifies reaction products by mass rather than by fluorescence, which is what allows a single reaction to carry up to 50 variants without the channel limit of a real-time PCR instrument.
Throughput
In the 96-well format the system holds two microtitre plates and two SpectroCHIP CPM 96 arrays, and processes up to 8 plates in a full working day plus 2 overnight. A plate takes 90 minutes of instrument time, 160 minutes including hands-on work, of which 5 to 10 minutes is actual handling. In the 384-well format it holds two 384-well plates and two SpectroCHIP CPM 384 arrays, processes up to 6 plates a day plus 2 overnight, and a plate takes about 7 hours. One 96 array carries up to 4,800 variants, one 384 array up to 19,200.
Applications
Genotyping of single nucleotide variants, insertions, deletions and translocations, copy number analysis, and methylation analysis in the EpiTyper workflow. Panels available on the platform include the VeriDose pharmacogenetics panels and the Hemo ID blood group panel. Interpretation software is offered by third parties through the MassARRAY Insights partner programme.
Regulatory status
The manufacturer states that all MassARRAY products are for research use only and not for use in diagnostic procedures, with the exception of the MassARRAY Dx System, which is CE-IVD.
The components that typically come with the Agena Bioscience MassARRAY® System include:
1. MassARRAY Analyzer: This is the core instrument that performs mass spectrometry-based analysis. It measures the mass of ions to determine the composition of nucleotides in DNA samples.
2. SpectroCHIP Array: This is a silicon chip used as a sample carrier for the MassARRAY system. It holds the samples and reagents in tiny wells, and it’s designed to be analyzed by the MassARRAY Analyzer.
3. SpectroCHIP Dispenser: This component is used to load samples and reagents onto the SpectroCHIP Array.
4. MassARRAY Typer Software: This is the software that controls the MassARRAY system, operates the instrument, and analyzes the data generated from the mass spectrometry analysis.
5. Assay Design Software: This software assists in designing genotyping or mutation detection assays that can be run on the MassARRAY platform.
6. Reagents and Consumables: Various reagents, buffers, and consumables are provided by Agena Bioscience to prepare samples, run the assays, and perform the mass spectrometry analysis.
7. Sample Preparation Equipment (optional): Depending on the specific application, you might also need additional equipment for DNA extraction, purification, and quantification prior to running the assays on the MassARRAY system.
