Why nucleosome-bound
Cell-free DNA in plasma exists as nucleosome-bound fragments, which is what gives it its characteristic length distribution around 150 bp. This material retains that structure, with a fragment size of 137 to 167 bp, so it behaves like patient cfDNA in extraction and in library preparation, whereas sheared DNA does not.
Applications
The manufacturer names four: spike-in experiments; use as a control with a very low noise background in workflow verification and validation; validation and development of sequencing and PCR protocols with regard to determining the limit of blank; and assessing the performance of an NGS pipeline. The low error rate is what makes the last two possible: at the level of minimal residual disease testing, the background of the reference material sets the floor of what the assay can distinguish.
Composition and storage
The material derives from the BDXXP4 lymphoblastoid wild-type cell line and is supplied in TE buffer at pH 8.0. Store at 2 to 8 °C on arrival and do not freeze. Production from another cell line is possible on request.
