Real-Time PCR Kit for Swine Flu

About Real-Time PCR Kit for Swine Flu

Swine Flu is a global public health concern, especially in immunocompromised populations. The TRUPCR® H1N1 (Swine Flu) Detection Real-Time PCR Kit provides fast, sensitive, and CDC-licensed detection of Type A Influenza viruses, including Pandemic Influenza A and the Pandemic H1N1 subtype. It enables real-time identification of viral RNA in respiratory specimens and viral cultures, facilitating timely clinical decisions.

The kit includes an internal positive control (RNaseP gene) to verify sample integrity and prevent false-negative results. It employs four parallel reactions in a single tube to simultaneously detect pandemic H1, pandemic Influenza A, general Influenza A, and the internal control—all through a single fluorescent channel.

With hot-start technology for increased specificity, the TRUPCR® kit is designed for use in molecular diagnostics, virology labs, and clinical microbiology. The workflow is streamlined, and the kit is validated across various real-time PCR platforms, making it an essential tool for swine flu-targeted surveillance and outbreak response.

What is in the Swine Flu Detection Kit

The TRUPCR® Swine Flu Detection Kit is a complete real-time PCR solution for detecting pandemic influenza strains. Each kit includes:

  • Primer and probe mix targeting four genes:
    • Pandemic H1 Influenza virus (H1N1)
    • Pandemic Influenza A virus (InfA pdm)
    • Type A Influenza virus (InfA)
    • Human RNaseP as internal control
  • One-tube multiplex reaction system (all in FAM channel)
  • Ready-to-use reagents and hot-start Taq polymerase
  • Compatible with viral RNA extracted via TRUPCR® Viral RNA Extraction Kit

This configuration ensures a streamlined, high-confidence workflow for detecting Swine Flu and related viral threats.

Role of Targeted Influenza Viruses in Disease

The 2009 outbreak of Swine Flu, caused by a novel H1N1 strain of Influenza A, spread rapidly and was declared a pandemic by WHO. The virus is a triple-reassortant with genes from swine, avian, and human influenza strains. Since then, H1N1-positive cases have been reported globally, with India experiencing major outbreaks between 2014 and 2015.

  • Type A Influenza virus (InfA): Encompasses all known seasonal and pandemic strains.
  • Pandemic Influenza A virus (InfA pdm): Indicates specific circulating pandemic variants.
  • Pandemic H1N1 virus (H1N1 pdm): The specific Swine Flu subtype responsible for high mortality during pandemics.

RNaseP, the internal control, is critical for verifying the presence of human nucleic acids and ensuring the accuracy of negative results.

About the TRUPCR® H1N1 Detection Kit

The TRUPCR® H1N1 (Swine Flu) Detection Kit is built on CDC-validated assay design, using real-time PCR for fast and reliable detection of pandemic and seasonal influenza. The kit is ideal for:

  • Early detection of Swine Flu-positive respiratory cases
  • Surveillance programs during influenza season
  • Outbreak management in clinical and public health settings
  • Confirmatory diagnostics in virology and molecular labs

Its one-step workflow reduces hands-on time, while its validated performance ensures confidence in every result.

Compatible platforms include Applied Biosystems 7300/7500, AriaMx, CFX96™, QuantStudio™ 3/5, Rotor-Gene Q, and StepOne™ systems.

Principle and Workflow

The kit employs a TaqMan®-based real-time PCR methodology with fluorogenic probes. Here’s how it works:

  1. RNA Extraction: RNA is isolated from respiratory specimens using validated kits.
  2. Reaction Setup: A one-tube mix includes primers, probes, enzymes, and internal control.
  3. Amplification: During each PCR cycle, the Taq polymerase cleaves the probe, separating the dye from the quencher.
  4. Fluorescence Detection: Increase in signal is measured in the FAM channel.

This Swine Flu-targeted multiplex system ensures sensitive, specific detection with minimal risk of cross-reactivity or inhibition.

Clinical Application

Swine Flu remains a threat, particularly in high-risk groups such as the elderly, immunocompromised, and healthcare workers. The TRUPCR® kit supports:

  • Rapid triage and diagnosis of flu-like illness during outbreaks
  • Hospital infection control and cohorting based on H1N1 status
  • Monitoring seasonal trends of pandemic and type A influenza strains
  • Public health screening in regions with known viral spread

By detecting pandemic H1N1, Influenza A, and including an internal control, the TRUPCR® kit enhances diagnostic accuracy and supports early intervention.
