The Viennalab IL28B RealFast™ Assay is a reliable and efficient way to predict the success of antiviral therapy and sustained virologic response (SVR) in HCV-infected patients. The assay accurately predicts the likelihood of HCV clearance by genotyping the functional variant rs368234815 TT>ΔG, which is the strongest known host factor for predicting HCV clearance.

Direct-acting antivirals and pegylated Interferon/Ribavirin regimens are commonly used for the treatment of HCV infection, but SVR is dependent on various factors, including virus genotype and patients’ age, sex, ethnicity, and genetic variants associated with antiviral activity. Genotyping of rs368234815 TT>ΔG using the Viennalab assay helps to predict spontaneous and treatment-induced viral clearance in patients, independent of their ethnicity.

Components of the IL28B PCR Assay
  1. DNA Extraction Kit: A kit containing all the reagents necessary for the extraction of genomic DNA from a blood or saliva sample, including DNA extraction buffer, proteinase K, and magnetic beads.
  2. PCR Mix: A ready-to-use mix containing all the reagents necessary for the PCR amplification of the IL28B gene region of interest.
  3. IL28B Primer Mix: A mix of primers specific for the IL28B gene region of interest.
  4. Probe Mix: A mix of probes specific for the detection of SNPs in the IL28B gene, including the rs12979860 and rs8099917 variants.
  5. Control DNA: A synthetic DNA that is added to the sample as an internal control to ensure that the assay is working correctly.
  6. DNase/RNase-free Water: Water that has been treated to remove contaminating DNases and RNases, which can degrade the extracted DNA.

 

The kit also includes instructions for use, as well as a protocol for the detection of SNPs in the IL28B gene using real-time PCR.
