CYP2D6 Stripassay

Short description

The Viennalab CYP2D6 PCR Kit is a powerful solution for accurate and efficient genotyping of the CYP2D6 gene, which plays a crucial role in the metabolism of about 25% of all drugs used in the clinic. It covers the most clinically relevant genetic variants and copy number changes associated with poor, intermediate, or ultrarapid CYP2D6 metabolizer status.

The kit allows for informed drug prescribing recommendations and dosage adjustments, reducing the risk of adverse events or lowered treatment efficacy. Clinical guidelines are listed on PharmGKB, and the PGX-CYP2D6 XL StripAssay® in combination with the CYP2D6 RealFastTM CNV Assay facilitates the identification of patients with altered CYP2D6 enzyme function.

Product highlights

  • CYP2D6 is involved in the metabolism of approximately 25% of drugs
  • CYP2D6 genetic variations include all alleles
  • Enables the identification of patients with altered CYP2D6 enzyme function

Specifications of the CYP2D6 Stripassay

Introducing the Viennalab CYP2C9 PCR Kit – the ultimate solution for detecting the most common variations in the CYP2D6 gene, which is involved in the metabolism of about 25% of all drugs used in the clinic, including tamoxifen, opiates, antidepressants, and antipsychotics. With the powerful combination of the PGX-CYP2D6 XL StripAssay® and the CYP2D6 RealFastTM CNV Assay, you can easily and accurately detect genetic variants and copy number changes in Caucasians, providing invaluable information to facilitate medication choice and dosage adjustment.

CYP2D6 loss-of-function, reduced function, and increased function alleles, including *3 – *8, *11, *12, *15, *40, *58, *114, *9, *10, *14, *17, *29, *41, *1xN, *2xN, *35xN (N= copy number), contribute to the inter-individual variability in drug response. The clinical guidelines listed on PharmGKB enable informed drug prescription recommendations.

With approximately 6% of Europeans being poor, 7% intermediate, and 3% ultrarapid metabolizers, it is crucial to identify patients with altered CYP2D6 enzyme function. The PGX-CYP2D6 XL StripAssay® in combination with the CYP2D6 RealFastTM CNV Assay allows for the identification of these patients, facilitating medication choice and dosage adjustment to reduce the risk of adverse events or lowered treatment efficacy.

Components of the CYP2D6 Stripassay
  1. DNA Extraction Kit: A kit containing all the reagents necessary for the extraction of genomic DNA from a blood or saliva sample, including DNA extraction buffer, proteinase K, and magnetic beads.
  2. StripAssay: A test strip containing immobilized probes specific for the detection of genetic polymorphisms in the CYP2D6 gene, including alleles *2, *3, *4, *5, *6, *7, *8, *9, *10, *17, and *41.
  3. Conjugate: A solution containing probes labeled with biotin that bind to the immobilized probes on the StripAssay.
  4. Enzyme Conjugate: A solution containing streptavidin conjugated to horseradish peroxidase (HRP) that binds to the biotin-labeled probes on the StripAssay.
  5. Substrate: A solution containing a chromogenic substrate for HRP that produces a color reaction upon cleavage by the enzyme.
  6. Stop Solution: A solution used to stop the enzymatic reaction and stabilize the color reaction.
  7. Control DNA: A synthetic DNA that is added to the sample as an internal control to ensure that the assay is working correctly.
  8. DNase/RNase-free Water: Water that has been treated to remove contaminating DNases and RNases, which can degrade the extracted DNA.

The kit also includes instructions for use, as well as a protocol for the detection of genetic polymorphisms in the CYP2D6 gene using the StripAssay format.
